Surface plasmon resonance (SPR) is a phenomenon occurring at metal surfaces(typically gold and silver) when an incident light beam strikes the surface at a particular angle.Depending on the thickness of a molecular layer at the metal surface,the SPR phenomenon results in a graded reduction in intensity of the reflected light.Biomedical applications take advantage of the exquisite sensitivity of SPR to the refractive index of the medium next to the metal surface, which makes it possible to measure accurately the adsorption of molecul ....etc

At an interface between two transparent media of different refractive index (glass and water), light coming from the side of higher refractive index is partly reflected and partly refracted. Above a certain critical angle of incidence, no light is refracted across the interface, and total internal reflection is observed. While incident light is totally reflected the electromagnetic field component penetrates a short (tens of nanometers) distance into a medium of a lower refractive index creating an exponentially detenuating evanescent wave. • Since its first observation by Wood in 1902 ,the physical phenomenon of surface plasmon resonance (SPR) has found its way into practical applications in sensitive detectors, capable of detecting sub-monomolecular coverage.
