Segmentation using watershed transformation works best if you can identify or "tag" foreground objects and background locations. Segmentation of marker-controlled basins follows this basic procedure:
1. Calculate a segmentation function. This is an image whose dark regions are the objects you are trying to segment.
2. Calculate the markers in the foreground. These are connected blobs of pixels within each of the objects.
3. Calculate the background markers. These are pixels which are not part of any object.
4. Modify the targeting feature so that it only has minima in the foreground and background locations.
5. Calculate the transformation of the basin of the modified segmentation function.
