The regeneration of new species and the improvement of cultivation techniques opened new horizons for practical breeding in a series of crops. The importance of protoplast sources and inclusion systems is discussed. The study of the reactive effects of oxygen species and DNA condensation, together with careful monitoring of phytohormones, are, in our opinion, the most promising research topics in the search for the rationalization of regeneration Of protoplasts. The following summarizes the progress of fusion and fragmentation. Genomic, transcriptomic and proteomic studies have led to a better understanding of fundamental processes such as cell wall formation, cell development and chromosomal rearrangement in fusion products, whether or not obtained after irradiation. Advanced methods of molecular screening of both the genome and the cytoplasm facilitate the efficient screening of symmetric and asymmetric fusion products. We expect emerging technologies such as GISH, high resolution fusion and next generation sequencing to make important contributions to our vision of genome creation and stabilization, especially after asymmetric hybridization. Finally, the agricultural valorisation of somatic hybridization is demonstrated by enumerating the recent introgression of various traits in a series of commercial crops.
